High-Capacity Chromatography Resin for Antibody and Antibody Fragment Capture
The newly developed Protein L affinity resin combines the sturdy TOYOPEARL polymer matrix with an engineered recombinant Protein L ligand. TOYOPEARL AF-r Protein L-650F demonstrates dynamic binding capacities (DBCs) exceeding 38 g/L for Fabs, nearly double the capacity of other commercially available Protein L resins.
Protein L exhibits a strong affinity for the variable region of an antibody's kappa light chain. Hence, TOYOPEARL AF-r Protein L-650F is suitable for purifying a wide range of antibody-related molecules that do not bind to Protein A. Examples include antibody fragments (Fabs), domain antibodies (Dabs), single-chain variable fragments (scFv), as well as IgM and IgA.
The recombinant ligand, fused with the well-established methacrylic polymer backbone of TOYOPEARL media, has been engineered to maintain capacity even after repeated exposure to mild alkaline solutions. Its multipoint attachment to the TOYOPEARL matrix further enhances its chemical stability. Additionally, thanks to its rigid polymer matrix, TOYOPEARL AF-rProtein L-650F offers excellent pressure flow characteristics.
